Preliminary Justification : Staphylococcus aureus stands out as a relevant human pathogen, because it is responsible for several infections and has a high infectious potential. Asymptomatic nasal and hand carriers of Staphylococcus aureus may act as disseminators of the microorganism to a population susceptible to colonization both in community and nosocomial environments. One of the important characteristic of Staphylococcus aureus is the ability to acquire mechanisms of resistance to antibiotics and the indiscriminate use of antibiotics can result in resistant strains. Therefore, the treatment of staphylococcal infections has become more challenging with the emergence of methicillin-resistant Staphylococcus aureus (MRSA), which are often also multi-drug resistant. Nursing students are at a higher risk of colonization by Staphylococcus aureus due to increased exposure to this organism, in addition to potential disseminators of these microorganisms in their care activities in hospital as well as into the community. Identifying and treating MRSA carriers may be effective in reducing both the incidence of nosocomial infections due to this organism and the risk of several infections in colonized health care workers, thus preventing the spread of MRSA to community. The aim of this study is to detect nasal and hands carriage of Staphylococcus aureus among nursing students from University of Nursing, Yangon. Moreover, antimicrobial susceptibility patterns will also test in order to establish the in vitro activities of antimicrobial agents against the Staphylococcus aureus isolates, which include methicillin-resistance Staphylococcus aureus (MRSA). Therefore, the result of this study will help to evaluate the possibility of spreading Staphylococcus aureus as well as MRSA among nursing students, to guide antimicrobial agents against MRSA and to educate students in order to prevent them from becoming nasal and hand carriers of MRSA.
 
Preliminary Aim : To study nasal and hands carriage of Staphylococcus aureus among nursing students from University of Nursing, Yangon, Myanmar
 
Preliminary Objectives : (1) To identify Staphylococcus aureus carriage in the anterior nares and on the the hands of nursing students by VITEK 2 Compact System (2) To determine the antimicrobial susceptibility patterns of Staphylococcus aureus isolates by VITEK 2 Compact System (3) To detect methicillin-resistant among Staphylococcus aureus isolates by cefoxitin disk diffusion test
 
Preliminary Sample Size : 220 samples of nasal and hand swab each from nursing students

Preliminary Study Method : 220 samples of nasal and hand swab each from nursing students at University of Nursing, Yangon will be collected from study population. One nasal swab and one hand swab each will be collected from nursing students. The nasal swab will be taken according to the guidelines laid down by CDC. The swabs will be withdrawn and kept in Stuart’s transport medium. Hand swabs will be taken from the participants with the sterile cotton wool swab stick from areas in between fingers, palm and wrist of both hand and placed into Stuart transport medium. Before taking hand swabs, hand washing of participants will be done with soap and water and then dried in air, not with antiseptic agent. Swabs collected from the nasal cavity and hand will be directly plated on nutrient agar and blood agar plates and incubated at 37 ℃ for 24 hours. After incubation, colonial morphology and pigmentation on nutrient agar and blood agar plates and any zone of haemolysis on blood agar will be identified. Gram’s stained smear of the isolate will be examined under oil immersion lens of the ordinary light microscope. The colonies of staphylococci will be collected from blood agar and the VITEK 2 compact system will be used for species identification and antimicrobial susceptibility testing of each isolate. Then, a suspension of Staphylococcus aureus isolates will be tested with cefoxitin 30 µg disc on Mueller-Hinton agar plate as a phenotypic confirmation of methicillin-resistance among Staphylococcus aureus isolates (≤21 mm = mecA positive, ≥22 mm = mecA negative).

Preliminary Research Finding : Data collected will be checked for completness, errors and inconsistencies before data entry. Data entry will be done by Microsoft Office Excel 2010 and data analysis will be done by Statistical Package for Social Sciences (SPSS) version 18.0.
